Metabolism and PK of these two constituents of kratom, and several relevant studies in rodents and in human plasma are discussed. It appears that a human-specific metabolite of 7-HMG is formed within people. The ability of these two constituents to cross the blood-brain barrier and their possible role in analgesic effects are explored. CYP enzymes and kratom’s interactions with central nervous system (CNS) drugs are relevant.
